Grenouille, having seen no human affection or experienced any of the values we inherit from human interaction such as love and compassion, find it difficult to distinguish right from wrong in his actions. Another text that has enhanced my understanding of how the concept of the outsider can be portrayed is the short film The Story of Bubbleboy. The film was produced in 2006 for the annual Australian tropfest short film festival. It was directed by Sean Ashcroft, produced by Kristy Fransen and stars Andrew Supanz. The Story of Bubbleboy is a film about a man named Bubbleboy who is trapped in the past and redeemed by the future. Bubbleboy is a sad outsider who lives alone due to a fanatic phobia of pointed and sharp objects. He is so afraid that he results to wrapping everything, including himself, in bubble-wrap. He never ventures from his house because the outside world is too terrifying and he cannot comprehend his fears. The film tells the story of his self-imposed exile from the world and how one day, driven by dwindling supplies he ventures out. Bubbleboy, while growing more confident as his adventure draws to an end, is ironically stabbed in an ally way. However, having been stripped from his protective bubbles in his last moments, Bubbleboy finds salvation and feels connected with the world. Because of Bubbleboys fear and self imposed isolation, he is regarded as an outsider by society. He completely avoids human contact and is scorned by the community. Bubbleboy longs for belonging but cannot comprehend his fears. A particular scene in the novel that conveys this idea is the beginning segment where we are introduced to Bubbleboys way of life and how it differs greatly from that of society. Here, filming techniques are used to show that Bubbleboy is an outsider. A flashing mirage of everyday pointy items is shown while a narrator explains Bubbleboys situation. There is then a long shot of Bubbleboy wrapped from head to toe in bubble-wrap huddling in a corner of his home. This initiates the idea that he is definitely an outsider and is uncomfortable with his surroundings. Close-ups are used to show the trepidation on Bubbleboys face to emphasize this point. A panning shot is then used to show how every household item has been covered in bubble-wrap, emphasizing the intensity of the problem. A birds eye view is also used here of Bubbleboy moving uncomfortably through his home; this suggests that he is a victim and is clearly suffering from his phobia. To show how Bubbleboys fears cause him to be perceived as an outsider by the community is the use of dramatic contrast between the outside world and his own. A view from the inside of the house looking out into the street brings a striking contrast of lively colors. When this is compared to the insides of Bubbleboys house, which is mostly colored in grays, the fact that he is an outsider is emphasized. Two young girls ride past the house and stop mocking Bubbleboys strange appearance from the window. Bubble boy then shuts his curtain and disappears. The use of the window here also symbolizes the physical barrier between himself and society. Contrast is used in a further scene in the film when illustrating the differences of Bubbleboys lifestyle to that of people his own age. Here, Bubbleboy is seen looking longingly into the window of a house where a lively and entertaining dinner party is taking place. A camera angle is used to show how Bubbleboy is looking up into the scene creating a sense of his inferiority and isolation. There are then a series of flashing images back and forth between the vibrant and colorful party life, with Bubbleboys lonesome isolation, again reinforcing this idea. When Bubbleboy finally faces his fears and leaves the house, more techniques are used to show how new and trepidating this is for him due to his experience as an outsider. For example when he opens the door into the outside world, a blinding and foggy light is emitted suggesting the unknown. Once outside the house, Bubbleboy again contrasts greatly with his surroundings as his general appearance is confronting and obviously very odd. Close-ups are used to show the looks of mockery and confusion on the towns peoples faces to convey this idea. The attitudes towards Bubbleboy as an outsider may be seen to change towards the end of the film. When he is ironically stabbed on his way home from his healing journey his is stripped of his bubble-wrap protection. It is here when society can accept him as his true self is revealed. This is shown by a woman rushing to the aid of injured Bubbleboy, truly concerned for his wellbeing. Bubbleboys perceptions of himself as an outsider can also be seen to change. A narrators voice is used to explain that at that point, Bubbleboy finds salvation, and for the first time feels connected to the world once outside his shell. There are not many similarities to the play Othello as the short film is such a vastly different text. Though it is clear that both characters; Bubbleboy and Othello are perceived as outsiders by society. However, it is Othellos unforeseen and irrational actions that confirm his status as an outsider. While it is Bubbleboys out of the ordinary actions that enable him to be accepted into society. Also, Bubbleboy is portrayed to be a victim of the imposing views of society, as he is a lonely and lost character, but on the other hand Othello victimizes society itself when he undergoes the actions that make him an outsider. Evidently, the three texts; Othello, Perfume and The Story of Bubbleboy have all increased my understanding of the outsider as they illustrate a vast use of techniques and concepts that make a person an outsider in society.

The topic has been considered a controversial one in terms of its definition and its definitive features. In a recent revision of the concept of famine, â€Å"Poverty and Famines,† Amartya Sen retains part of classical vision on famine offered by Malthus, distinguishing â€Å"regular starvation,† which is a â€Å"normal feature in many parts of the world,† from â€Å"violent  outbursts of famine,† a â€Å"particularly virulent form of [starvation] causing widespread death† (Sen, 38-39). USAID defines famine as â€Å"a catastrophic food crisis that results in widespread acute malnutrition and mass mortality† (USAID, 2002).   Proper definition of famine matters not only in terms of labeling an event after the fact, but also in terms of how humanitarian organizations and governments respond to crises as they are happening. Maxwell points out that this is in large part because of the emotional weight the term â€Å"famine† has come to carry (Maxwell, 49). Humanitarian workers spent a considerable amount of time arguing about whether or not to call the 2002-2003 crisis in Ethiopia, ostensibly affecting over 13 million people, a famine. Calling it a famine would have stepped up the international response, but it might also be perceived as â€Å"crying wolf,† which would have a detrimental effect on organizations’ abilities to obtain resources for emergency responses in the long run. Aid agencies want to avoid using the term â€Å"famine† too often because they worry about â€Å"compassion fatigue† or â€Å"donor fatigue† – essentially that donors will be less likely to support emergency efforts if there are too many emergencies. There are also political implications for using the term â€Å"famine,† as can be seen in the case of the 2005 crisis in Niger, which President Mamadou Tandja insisted was a fabrication of relief agencies to obtain more funding (Sengupta, 2005). Aid agencies likewise were reluctant to apply the term famine, and referred instead to â€Å"pockets of severe malnutrition,† in part because they didn’t want to alienate Tandja (Sengupta, 2005). The Nestles objectives of outsourcing are cost reduction and reduce task which is not important, so the company can focus on their core processes.(Nestle, 2012) What Nestle uses to determine that the company should outsource or keep it in-house are how the processes or operations important and capacity of its own according McIvors theory (2008).McIvors theory (2008) explains that if any processes are the core processes of the company and the company is less potential, the company should consider that whether the processes can be copied. If the processes can be easy to replicate, the company should keep in-house, on the other hand, if they are difficult to copy, the company can outsource. David Walker (2013), Nestle customer services manager explains that the results of OmPrompt system were used to compare with the existing manual order entry processes. Logistics and supply chain outsourcing Nestle outsources Geodis to manage its logistics and supply chain in France. Nestle target is reduced transportation cost which is about 10-15% of finished product (ElAmin, 2006).   In the beginning, Nestle assigned Geodis to study its supply chain flow to identify the area which will be improved. After six months, Geodis sent the result and recommendation solutions to Nestle. After considering the result, Nestle appointed Geodis as its logistics provider for three years for all it transportation flow. In return Geodis has made performance guarantees relating to quality, optimisation and total cost reduction. Geodis will be looking to improve Nestles transport and supply chain flows, logistics, management of service providers and invoice checking, among other factors (ElAmin, 2006)  . Following the agreement Geodis have to manage production flow for six Nestle France plants. The process made up of 17,000 transport orders per year from 182 suppliers and 250 contries (ElAmin, 2006). In term of contracting and relationship, we can notice that Nestle used a short term contract in the beginning to ensure the outsourcing company that can respond Nestles need by assigning the outsourcing company as a third party to identify the Nestle problems and solutions. This step shows relationship between Nestle and Geodis that they are unfamiliar, so Nestle had to measure Geodis performance before they offer larger jobs and long term contract. When Geodis can do the job well, so Nestle was satisfied the result and offer the long term contract and shift relationship with Geodis from 3PL to 4PL to manage its all supply chain. Task 2 Nestles Challenges and Solution Corporate social responsibility  (CSR)  and ethic challenge From Nestle is the world company, so ethic and responsibility for social and environment are important. For example, in February 2012 Nestle joined Fair Labour Association to create sustainable supply chain and prevent child labour on cocoa supply chain in Ivory Coast (CNN, 2012). As a result, when Nestle outsources any company, Nestle should identify this standard to the third party or set the criteria in the contract or agreement to the third party following the same way that Nestle responds to social otherwise Nestle might force problems from what outsource do. For instance, when independent investigator examines Nestles cocoa processes in Ivory Coast, they found that Nestles outsourcing using child labours to harvest raw cocoa for a long time (CNN, 2012, Thetimes100, n.d.B). The good case study which Nestle can use to be example is IKEA, home furnishing company focusing on sustainability and social responsibility from all processes of supply chain, so IKEA set its vision that to create a better everyday life for many people. IKEA uses suppliers to provide raw material in primary stage to produce products. IKEA checks the environmental impacts and keep an eye on responding to the social at suppliers. Furthermore, IKEA sets conditions such as no allowing child labour, emissions reduction, to the suppliers before sign the contract. Every condition help to develop sustainability and social. To control suppliers, IKEA also analyses documents and records and visit on-site on occasions to make sure its suppliers are still following the agreements. IKEA works with UNICEF to develop community program in India to prevent child and raise awareness (Thetimes100, n.d.A). In addition, Nestle can adapt the result from launch cocoa plan Indonesia, the worlds third largest coffee producing country (CNN, 2012). Nestle assigns research and develop team in France to study about how to improve productivity, and then send their staff to help the farmers improve lives. Nestlà © and partners to train farmers provide plant expertise and support supply chain transparency. It also aims to improve livelihoods by focusing on projects related to nutrition education, water and rural development (Badasha,K, 2012). Nestle can use this concept to launch in Ivory Coast as well. It can help the farmers improve effective and earn more money; on the other hand, Nestle will get more raw materials. It means win-win situation, which can cause sustainability to the supply chain. Supplier Selection Challenge In 2007 Nestle outsourced Barry Callebaut to manufacture its chocolate products while Hersheys and Cadbury which are chocolate companies also chose Barry Callebaut to produce their products. Until 2012 Nestle decided to stop outsourcing from any company and keep production at York where can produce 8000 tons of dark chocolate for Kit Kat (BarryCallebaut, 2012, Nieburg, 2012).   Before outsourcing should be carefully considered in many respects such as Nestles capacity, relationship between outsource company and competitors. When we consider the situation we can see that Nestle is large food company. Chocolate is the popular products and Nestle familiars with productive processes for a long time (Nestle, 2013). Producing chocolate can be the core process and strength of the company, so Nestles potential can support chocolate production by itself. Although outsourcing Barry Callebaut reduced Nestles tasks and boost Nestle performance up, Barrt Callebaut also is outsourced from Hershey and Cadbury which are Nestles competitors. As a result, Nestle should make sure that its important information will not be shared to competitors because many times competitors have close relationship with third party. When Nestle stop outsourcing from Barry Callebaut. Barry Callebaut use knowledge from Nestle to support competitors or create own products to challenge Nestles mar ket. In this case, Nestle should invest to develop its potential at the beginning to keep significant processes in-house or find effective companies which can rely on only Nestle and improve relationship with them, however, Nestle should study more about background of the outsource company including its customer or any company which they are supporting. Supply Chain Challenge Nestle establishes its huge logistics by using two largest UK distribution centre in York and Bardon in Leicestershire. In 2002 Nestle faced problems of over and under utilisation with two distribution centre because it sold off a critical part of Premier food business although Nestle outsourced TDG, one of the greatest supply chain providers to control operation in two hubs. Nestle assigns York to respond Nestlà ©s confectionery products, the rest of products including food section are handled by Bardon. After selling food part, it made Bardon be under utilisation, while York was supporting confectionery products over utilisation. It created unbalancing usage (Total-logistics. n.d.).   Re-engineering to solve the problem One of the possible solutions mitigate under utilisation affect of the food sectors by analyse and redesign function of two distribution centres to balance usage and improve to be regional centres by changing the existing responsibility for each hub from the old pattern to mix products in each distribution centre. It means each distribution centres should be assigned to carry the full range of products, so over utilisation from confectionery products could be shared to under utilisation in Bardon. In addition, it could reduce lead time in supply chain when shift distribution centres to be regional centres because in several cases Nestle negotiate with its customers to pick up the products at Nestles site (Total-logistics. n.d.)  , so Nestle could arrange the shortest distance between customers and its regional centres, on the other hand Nestle can also arrange its transportation and regional centres to suite the customers in each area. It makes Nestle increase responsiveness and save cost al well. However, the two hubs have to be linked in term of information system, processes and transportation. They should be generated by Nestle and TDG cooperation. For example, TDG at Bardon and also covered the AUTOSTORE warehouse management system (WMS) running all product movements at both sites. All of which would need to fit hand in glove with Nestlà ©s SAP enterprise resource planning (ERP) system located at the companys Croydon headquarters (Total-logistics. n.d.).   Conclusion As we can see, many things from outsourcing are support Nestle business such as IT, logistic and supply chain etc. The two main benefits from outsourcing are cost reduction and unnecessary operational reduction to help outsourcer can focus on the core operation of the business. As a result, many factors should be considered before outsource such as core competitive and potential of the company not only outsourcing but also the outsourcer to get the most value from outsourcing and not generate the bad impact to the company. Some perspectives from Nestle reflect and demonstrate outsourcing strategies and challenge from this report and can be studies more in the future.

Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) in the downregulation of Bamboo mosaic virus and its associated satellite RNA Replication :: Biology, RNA

The association of host proteins with viral replicase complexes has been demonstrated in a number of plus-strand RNA viruses (1, 24), including the Bamboo mosaic virus (BaMV). In BaMV it has been reported that chloroplast phosphoglycerate kinase (PGK) (25) and HSP90 (Huang et al., unpublished data) are required for the efficient accumulation of BaMV; where as the identity of the additional factors associated within the BaMV RdRp complex, and the proteins involved in satBaMV RNA replication are not yet been acknowledged. This study identified a host metabolic enzyme namely GAPDH, that interact to negatively regulate the Bamboo mosaic virus (BaMV) and its associated satellite RNA accumulation. The RNA binding properties of GAPDH has already been documented for a number of viruses (9, 14, 29, 41, 53, 56). However, the interaction of GAPDH protein with different viral RNAs results in a functionally different mode of regulation on viral replication and translation. For instance: GAPDH int eracts with the JEV NS5 protein indirectly by binding with 3†²-ends of JEV, resulting in virus-induced redistribution of GAPDH to control the early stage of JEV replication/translation (53). GAPDH plays a major functional role in the replication of tombusviruses through the retention of the viral minus-strand RNA template in the replication complex in order to promote asymmetric RNA synthesis (48). In contrast, GAPDH inhibits viral replication in the interaction with other viruses. For example: Silencing GAPDH increases TGEV infection by 2-3 times, demonstrating the anti-TGEV activity of this protein (14). Binding GAPDH to the HAV RNA suppresses cap-independent translation due the destabilization of the secondary structure of RNA (55). In our study, a downregulation of GAPDH-C led to a 2 to 3-fold increase in the replication BaMV and satBaMV RNA, indicating that GAPDH-C has an inhibitory effect on BaMV and satBaMV infection. In addition, an increase in BaMV-GFP was observed on inoculated leaves in GAPDH-C silenced N. benthamiana, revealing that GAPDH-C functions in the early stages while the virus is establishing a successful infection of the primary invaded cells.
